Abstract

Major depressive disorder (MDD) is the most prevalent mental illness, which is a chronic, recurrent, and even life-threatening mental disease. Despite the wide clinical
use, the current antidepressant medications, including selective serotonin reuptake inhibitors (SSRIs) have been largely limited to treat this disease timely and
effectively without unnecessary side effects. More seriously, our understanding of the anti-depressive mechanisms of those drugs, is yet to be far beyond the elevation of
serotonin level in the brain. In particular, the innate complexities of the neural system, most likely due to the heterogeneity of neuronal subpopulations, have
hampered our approach to cell-type specific molecular analyses of the antidepressant actions in vivo. By combining transgenic and genomic approaches in
the rodent models, we characterized the role of a neuronal subtype for the drug actions in the hippocampus. With TRAP (translating-ribosome affinity purification)
approach, we further explored the cell type-specific transcriptional alteration, which may underlie adaptive changes of the neural circuits to chronic SSRI administration.
This study may contribute to better understanding of the molecular basis underlying the therapeutic or the unnecessary actions of the SSRIs and further to the global
effort to develop the advanced therapeutics for the MDD patients.
