Abstract

Mitochondria division is critical for maintenance and regulation of their function and distribution. At the contact site between endoplasmic reticulum and mitochondrial outer-membrane, actin filaments and constriction ring by dynamin-related protein 1 (Drp1) mediate mitochondrial constriction preceding the actual division. Recent reports have demonstrated that intra-mitochondrial physiological status, including oxidative phosphorylation, is also important for complete mitochondrial division as well as Drp1 action. However, how the intra-mitochondrial physiology contributes to efficient mitochondrial division is unclear. Here, we discovered spontaneous and repetitive constriction of mitochondrial inner-compartments (CoMIC) in neuronal processes, often associated with subsequent mitochondrial division. Interestingly, this event was dramatically augmented by blockade of Drp1, which appears predominantly on mitochondrial inner compartments. In addition, the CoMIC was facilitated by intra-mitochondrial potassium influx through mitochondrial big-conductance calcium-dependent potassium channel and accumulation of soluble short Opa1 by Oma1. Therefore, we provide novel evidence for the molecular mechanisms of intra-mitochondrial events for complete mitochondrial division.
