Abstract


Type I collagen, the most abundant protein in our bodies, fulfills diverse mechanical, biological and even optical functions in a variety of tissues. To accomplish these functions, sequence specific collagen peptides are assembled into tissue-specific, hierarchical architectures that are structured at all length scales from nanoscale to macroscale. When this sequence specific peptides and their assembly process are impaired, severe disease states emerge. Our understanding of collagen structures and functions, and our ability to mimic artificial collagen functions in vitro are therefore critical to achieving advancements in developmental biology, wound healing, biomechanics, tissue engineering, regenerative medicine, and the treatment of collagen-associated diseases.
In my seminar, I will introduce a new way to develop collagen-like peptide sequences through directed evolution process of phage display. Specifically, I will introduce construction of collagen-like peptide library and discovery of novel function collagen-like peptides. I will then demonstrate to utilize these peptides for targeted drug delivery for possible cancer therapeutics. Inaddition, I will show how to construct functional nanomaterials through synthesis of peptide amphiphiles with discovered peptides for soft and hard tissue engineering. Our approaches to develop collagen-like peptides and to create their functional materials will be very useful for the novel protein-based targeted drug vehicles and tissue engineering materials.
