Abstract


The process of RNA synthesis and splicing of cytokine genes is an essential step for activating effective host defense against microbial invaders, thus dysregulation of cytokine expression causes chronic infection conditions or inflammatory diseases. Here, we report that cellular nucleic acid-binding protein (CNBP) acts as a transcription factor that is required for activating the innate immune response. In particular, CNBP selectively bound to specific regulatory elements on the promoter region of inflammatory genes, leading to their maximal production. We also demonstrate that a subnuclear body called the interleukin-6 (IL-6) splicing activating compartment (InSAC) is a nuclear site of IL-6 RNA splicing and stability. Tat-activating regulatory DNA-binding protein-43 (TDP-43) functions as an InSAC scaffold that selectively associates with IL-6 RNAs in a sequence-specific manner. The interaction of TDP-43 with spliceosomal components from the Cajal body leads to the formation of InSAC. Thus, we propose that inflammatory RNA production by CNBP and InSAC represents a new process to control the quality of inflammatory cytokine gene expression and to maintain immune activation during infections.
