Abstract
Organelle contact sites are specialized intracellular zones called membrane contact sites (MCS), in which two distinct suborganelles are closely apposed in eukaryotic cells. The MCSs play pivotal roles in cellular processes such as cooperative lipid biosynthesis, ion homeostasis, and interorganellar trafficking of molecules.
The MCSs are formed through dynamic and direct interactions between proteins
that are located in two distinct subcompartments. In this talk, I will introduce structural and functional studies for the ER-mitochondria contact sites (ERMCS) in yeast and mammalian cells. In yeast, the ERMCS is mediated by protein complex called ERMES (the endoplasmic reticulum–mitochondria encounter structure). Based on findings revealed by my group, I will address the molecular mechanisms by which the ERMES complex is organized and functions in lipid trafficking at this region. In addition, based on the recent study, I will introduce a novel mammalian protein to form the ERMCS and facilitate lipid trafficking in higher eukaryotic cells.
