연제 : Transfer-RNA-derived small RNA: a novel regulator of mRNA translation and biomarkers of disease

Abstract

There are thousands of different transfer-RNA-derived small non-coding RNAs (tsRNAs) of 18-40 nucleotides in length in mammalian cells. 
We found that a LeuCAG3′tsRNA derived from 3′ end of tRNALeu-CAG regulates cell viability by altering the secondary structure of ribosomal protein S28 (RPS28) and S15 (RPS15) mRNAs during their translation, which are critical for ribosome biogenesis. 
High-throughput sequencing data from NCI (national cancer institute) cancer database suggest that this tsRNA might be highly expressed in human hepatocellular carcinoma and can be potential targets for cancer treatment. Systemic delivery of antisense oligonucleotides to LeuCAG3′tsRNA into mice bearing patient-derived orthotopic hepatocellular carcinomas induced apoptosis and significant growth retardation in patient tumors. Our results suggest that at least some of 3′tsRNAs regulate protein synthesis by a newly defined mRNA translation mechanism which is conserved between species and provide novel targets for cancer therapeutics.
